Vα24+Vβ11+NKT细胞亚型在多发性骨髓瘤的研究

摘要:多发性骨髓瘤(multiple myeloma,MM)是一种血液系统恶性肿瘤。有报道表明,自然杀伤T细胞(NKT,Natural killer T cells)极有可能参与MM的发生发展,成为治疗MM的新突破口。本研究通过收集MM病例组和健康对照组的骨髓和外周血样本,使用流式细胞术检测Vα24+Vβ11+NKT细胞亚型CD4+NKT、CD8+NKT、CD4+CD8+NKT、CD4-CD8-NKT细胞在MM患者和正常健康人中的表达水平,探讨Vα24+Vβ11+NKT细胞亚型与MM的相关性及临床意义,研究发现MM患者Vα24+Vβ11+NKT细胞亚型出现了比例失衡,MM肿瘤细胞中具有肿瘤免疫功能的CD8+NKT、CD4-CD8-NKT细胞比例下降,而具有免疫调节功能的CD4+NKT细胞比例上升,这种比例失衡可能是造成MM肿瘤细胞逃逸免疫监视的机制之一。本研究为MM的发病机制和治疗提供新思路。

Investigation on Vα24+Vβ11+NKT cells'subtypes in patients with multiple myeloma
Abstract:Multiple myeloma (MM) is one kind of hematologic malignancy. Study reported that Natural killer T (NKT) cells are very likely to be involved in the occurrence of MM, are also one of the breakthroughs for the treatment of MM. This study mainly analyzed Vα24+Vβ11+NKT cells' subtype expression in MM. The researchers collected the bone marrow and peripheral blood samples of MM patients and healthy controls, and detected the expression level of CD4+ NKT cells, CD8+ NKT cells, CD4+ CD8+ NKT cells and CD4- CD8- NKT cells in patients with MM and healthy people by flow cytometry. They found that compared with normal controls, Vα24+Vβ11+NKT subtype percentage in peripheral blood of MM patients were inconsistent, in which CD8+NKT and CD4-CD8-NKT cells decreased, while CD4+NKT cells increased which are associated with anti-tumor immunity and immune regulatory function respectively. This inconsistency may be one of the mechanisms of MM tumor cell escape from immune surveillance. This study provides a new way of the pathogenesis and treatment of MM.
