利用不同方法研究多糖和寡糖与半乳凝素-3的相互作用

摘要:本文利用两种直接结合法和三种竞争性抑制法,研究了多糖和寡糖与半乳凝素-3之间的相互作用。直接结合法包括:表面等离子共振法(SPR)和生物膜干涉法(BLI);竞争性抑制法包括:荧光偏振法(FP)、竞争性荧光免疫抑制法(cFLISA)和半乳凝素-3介导的红细胞凝集法(G3H)。实验结果表明,SPR和BLI法适用于检测多糖和半乳凝素-3之间的相互作用;FP和cFLISA法适用于检测寡糖和半乳凝素-3之间的相互作用,但是与探针的选择有很重要的关系;G3H法既适用于多糖,也适用于寡糖。实验结果对糖与蛋白质相互作用的研究提供了重要的参考方法。

Approaches used to evaluate the interaction between polysaccharide or oligosaccharide and galectin-3
Abstract:Two direct binding assays, surface plasmon resonance (SPR) and bio-layer interferometry (BLI)), and three competitive inhibiting assays, ?uorescence polarization (FP), competitive fluorescence-linked immunosorbent assays (cFLISA), galectin-3-mediated hemagglutination (G3H) were used to evaluate binding of pectic-derived polysaccharides and oligosaccharides to galectin-3. Results show that whereas SPR and BLI work well to assess polysaccharide galectin-3 binding, they provide less reliable and less consistent binding data with oligosaccharides. Results using FP and cFLISA assays depend greatly on the selection of the probe and work better with oligosaccharides than with polysaccharides. The G3H assay provided reliable and consistent results with both polysaccharides and oligosaccharides. The results are useful for studying the interaction between glycoconjugates and proteins.
