血红蛋白清道夫受体在间充质干细胞向血管内皮细胞分化过程中的分子机制研究

摘要:骨髓间充质干细胞(BMSC)分化为血管内皮细胞(VEC)这一特性,可以为治疗包括局部缺血或者糖尿病等多种疾病提供可能。然而骨髓间充质干细胞向血管内皮细胞的分化过程尚未研究清楚。最近的报道显示血红蛋白清道夫受体(CD163)的表达与血管生成存在紧密的联系。在本研究中,在骨髓间充质干细胞中过表达血红蛋白清道夫受体(CD163)可以上调内皮相关标志蛋白CD31,Flk-1,eNOS和VEcadherin的表达,而且可以有效提高细胞对LDL的吞噬能力,进一步证明了血红蛋白清道夫受体(CD163)位于Hmbox1的下游同时位于FGF-2的上游。这些数据表明血红蛋白清道夫受体(CD163)参与到了Hmbox1/CD163/FGF-2所介导的骨髓间充质干细胞向血管内皮细胞分化的信号通路中。本研究发现了一个新的信号通路,同时为进一步研究骨髓间充质干细胞向血管内皮细胞分化过程提供了新的靶点。

Molecular Mechanism Study on the Hemoglobin Scavenger Receptor in the Differentiation of Marrow Stromal Cells into Vascular Endothelial Cells
Abstract:Bone marrow stromal cell (BMSC) can differentiate into many kinds of cells, including vascular endothelial cell (VEC). Recently, more and more researches suggested that there is a relationship between the hemoglobin scavenger receptor (CD163) and angiogenesis. In this paper, the hemoglobin scavenger receptor (CD163) elevated the expression of the endothelial markers, improved the Alexa Fluor 488-acetylated-LDL uptake capacity, and promoted angiogenesis on Matrigel. Furthermore, during this differentiation, the expression of the hemoglobin scavenger receptor (CD163) which led an increasement of the fibroblast growth factor 2 (FGF-2) expression, was regulated by homeobox containing 1 (Hmbox1). Therefore, we demonstrated the Hmbox1/CD163/FGF-2 signal pathway in BMSC differentiation into VEC.
