基于试纸的比色法分析传感平台用于尿液中葡萄糖和蛋白的检测

摘要:我们针对尿样中的葡萄糖和蛋白质分别在纸上开发了可视化的比色检测的方法。在葡萄糖的检测中使用了葡萄糖氧化酶和辣根过氧化物酶双酶体系,这两种酶与2 ,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(ABTS)联用可以在纸上和葡萄糖水溶液发生显色反应。在蛋白质的检测体系中,我们将四溴酚蓝(TBPB)滴在试纸上,在有蛋白质的水溶液滴在纸上时,两者结合会引起颜色从黄到蓝的变化。最后利用软件分析了两种体系中的颜色变化并得到其检测范围,然后把这种方法应用于实际尿样中葡萄糖和蛋白的检测。

Paper-based colorimetric sensing for detecting glucose and protein in urine
Abstract:We developed two visual colorimetric assays for detecting glucose and protein in urine sample on the paper. For the glucose assay, glucose oxidase (GOX) catalyzed the oxidation of glucose to gluconic acid and hydrogen peroxide, which would oxidize ABTS to produce green-color product by the catalysis of horse radish peroxidase (HRP). For the protein assay, the ionized TBPB was added to the paper firstly, then the color changed from yellow to blue upon addition of protein. The color of paper was taken by smart phone and the images were further proceed with ImageJ to to carry out linear regression for glucose and protein. Finally, we analyze the glucose and protein in the urine samples to verify the method can be used to real sample test.
