草鱼 ghrelin 基因的克隆、组织表达及 ghrelin 和 obestain 蛋白对其摄食和生长激素分泌的影响

摘要:Ghrelin 和 obestatin 是由同一前体物质 preproghrelin 翻译后加工剪接等过程获得的两种脑肠肽。本实验利用同源克隆技术获得了草鱼 ghrelin 基因的编码区序列(CDS 序列),其 cDNA 序列全长 491 bp(GenBank 序列号no.KC432584),草鱼 ghrelin 基因包括 4 个外显子和 3 个内含子,与大多数硬骨鱼类的 ghrelin 基因结构相同。编码的 103 个氨基酸的 ghrelin 前体肽,经剪切加工后形成含有 19 个氨基酸的 ghrelin 成熟肽[GTS(O-n-octanoic acid) FLSPAQKPQGRRPPRV]和 22 个氨基酸的 obestatin-like 成熟肽[APFELSVSLSEAEYEKYGPVLQ-NH2],质谱分析表明 ghrelin 和 obestatin-like 成熟肽的分子量分别为 2205.57 Da 和 2455.75 Da。草鱼腹腔注射 ghrelin(100 ng g-1 BW)后摄食量显著性增加,同时注射 ghrelin(100 ng g-1 BW)与 obestatin-like 蛋白(25 ng g-1 BW)表现出抑制 ghrelin 引起的摄食量增加。腹腔内注射 ghrelin(100 ng g-1 BW)能显著性提高草鱼垂体 GH mRNA 表达水平以及血浆 GH 水平,腹腔注射 obestatin 蛋白(25 ng g-1 BW)也表现出抑制 ghrelin 引起的垂体 GH mRNA 表达水平以及血浆 GH 水平变化。实验结果表明,草鱼 ghrelin 能够提高垂体 GH mRNA 表达水平,刺激 GH 释放,促进摄食。腹腔注射 obestatin 蛋白部分消除 ghrelin 刺激草鱼摄食的作用,表明 obestatin 蛋白可能参与介导 ghrelin 对摄食的调节。

Molecular cloning, tissue distribution of grass carp ghrelin and effect of ghrelin and obestatin on its food intake and growth hormone secretion releasing
Abstract:Ghrelin and obestatin are two gastrointestinal peptides obtained by post-translational processing of a common precursor, preproghrelin. Nucleotide sequences of ghrelin gene were identified both at DNA and cDNA levels, using homology-based cloning techniques from grass carp (Ctenopharyngodon idellus). We have cloned the complete cDNA and genomic DNA sequences of grass carp ghrelin precursor. Full-length cDNA sequence of grass carp ghrelin precursor was 491 bp (GenBank no. KC432584), which encoded a peptide of 103 amino acid residues. The primary sequence of grass carp ghrelin was predicted to be a 19-amino acid peptide, GTS(O-n-octanoic acid)FLSPAQKPQGRRPPRV, of which molecular weights were 2205.57 Da, and a 22-amino acid peptide obestatin-like, APFELSVSLSEAEYEKYGPVLQ-NH2, of which molecular weights were 2455.75 Da. Intraperitoneal (IP) injection of ghrelin (100 ng g-1 BW) increased pituitary GH gene expression and plasma GH levels. IP injection of the fragment of obestatin (25 ng g-1 BW) decreased ghrelin-induced pituitary GH gene expression and plasma GH levels. Food intake was significantly increased by IP injection of ghrelin (100 ng g-1 BW), but not by ghrelin at 100 ng g-1 BW in combination with obestatin at 25 ng g-1 BW. The results indicated that ghrelin stimulated GH gene expression and GH release, ghrelin was involved in the peripheral modulation of the FI ( food intake) in grass carp. The IP administration of obestatin at 25 ng g-1 BW, partially blocked the ghrelin-induced feeding, it investigated the possible involvement of obestatin as mediators of the ghrelin stimulatory action on food intake.
