抗肿瘤药用植物内生真菌的分离及其代谢产物抗肿瘤活性研究

摘要:目的 随着发病率和病死率的不断增高,恶性肿瘤严重地威胁着人类的健康和生命,已成为一个严峻的公共卫生问题。由于临床抗肿瘤药物存在一定问题,寻找新型高效低毒的抗肿瘤新药或新药先导化合物具有重要意义。方法 采集长白山地区13种抗肿瘤药用植物,分离、鉴定植物内生真菌;利用MTT法根据对Hela细胞的抑制率和IC50值,筛选出具有较高抗肿瘤活性的内生真菌;并观察抗肿瘤有效成分对Hela细胞的抑制作用。结果 从抗肿瘤药用植物中共分离获得17属108株内生真菌,其种类多样。其中4株植物内生真菌的次级代谢产物具有较高的抗肿瘤活性,对Hela细胞的抑制率≥85%,3株菌IC50值≤50μg/mL。抗肿瘤活性最高的内生真菌FI 29的抗肿瘤有效成分,对Hela细胞的抑制作用呈明显的浓度和时间依赖性。结论 抗肿瘤药用植物中内生真菌的种类具有丰富的多样性。通过筛选找到了对肿瘤细胞抑制作用较强的内生真菌有效成分,可为开发高效低毒的天然抗肿瘤新药或新药先导化合物提供有益资源。

Antitumor activity analysis of endophytic fungi isolated from antitumor medicinal plants
Abstract:Object With increasing incidence and mortality, cancer is the leading cause of death and is a major public health problem. And there are some problems of clinical antitumor drugs. Endophytic fungi have variety natural bioactive products, which is a potential source of new antitumor drugs or new lead compounds. Methods Thirteen species of antitumor medicinal plants in Changbai Mountain were collected. Endophytic fungi were isolated and identified according to morphology. By MTT assay based on inhibition rate and IC50 values on Hela cells, endophytic fungi with a higher antitumor activity were selected, and the effects of endophytic fungal active ingredient on Hela cells were observed. Results There are 17 genera and 108 strains of endophytic fungi were isolated from antitumor medicinal plants with species diversity. The secondary metabolites of 4 isolates had better antitumor activity, with the inhibition rate on Hela cells ≥85%, and 3isolates with IC50 values≤50μg/mL. The effects of active ingredients of endophytic fungi FI 29 with the highest antitumor activity on Hela cells showed an obviously concentration and time dependent manner. Conclusions The endophytic fungi isolated antitumor medicinal plants have variety diversity. The endophytic fungi active ingredient had a higher inhibitory effect on tumor cells is excavated in this study, which is useful resources for the research of natural and effective antitumor drugs or new drug lead compounds.
