药用植物内生真菌多样性及其代谢产物抗真菌活性研究

摘要:目的 近年来,真菌感染的发病率不断升高,而临床抗真菌药物存在一定问题。植物内生真菌可以产生种类丰富、结构新颖、天然低毒的生物活性物质,从中寻找新型抗真菌药或新药先导化合物具有巨大的开发潜力。方法 采集10种药用植物的叶或茎,分离植物内生真菌,利用形态学鉴定种类。采用液基微量稀释法检测次级代谢产物的抗真菌活性。结果 从药用植物中共分离获得18属84株内生真菌,其存在地域性差异和宿主差异。其中13株植物内生真菌的次级代谢产物抗真菌活性较好,其最低抑菌浓度(MIC)为25~100μg/mL。分离自石斛的内生真菌尖孢镰刀菌抗真菌活性最高,MIC为25μg/mL。结论 10种药用植物中内生真菌的种类和分布具有丰富的多样性。宿主植物对植物内生真菌的抗真菌活性存在一定影响。研究中发现的抗真菌活性较高的植物内生真菌可为寻找天然、高效的抗真菌新药或新药先导化合物提供较好的资源。

Diversity and antifungal activity of endophytic fungi isolated from medicinal plants
Abstract:Object In recent years, the incidence of fungal infection increased, and there are some problems of clinical antifungal drugs. Endophytic fungi were reported to produce variety, novel, and low toxic natural bioactive agents, which would be a potential source of new generation of antifungal agents or new lead compounds. Methods The leaves or stems of 10 species of medicinal plants were collected. Endophytic fungi were isolated from them and identified with morphological characteristics. Then, broth dilution antifungal susceptibility testing was done to detect the antifungal activity of secondary metabolites with these isolates. Results There are 18 genera and 84 strains of endophytic fungi were isolated from 10 species of medicinal plants, with the differences of regional and host. The secondary metabolites of 13 isolates had better antifungal activity, with the minimal inhibitory concentrations were 25~100μg/mL. The antifungal activity of endophytic fungous Fusarium oxysporum isolated from Dendrobium was highest, with MIC 25μg/mL. Conclusions Species and distribution of endophytic fungi isolated from 10 species of medicinal plants have variety diversity. The antifungal activity of endophytic fungi is affected by the host plants. The endophytic fungi with higher antifungal activity in the study are better resources of natural and effective antifungal agents or new lead compounds.
