酵母聚糖佐剂联合人类成纤维激活蛋白DNA疫苗的有效诱导小鼠抗肿瘤免疫应答

摘要:目的 观察酵母聚糖佐剂联合人类成纤维激活蛋白(FAP)DNA疫苗免疫BALB/c荷瘤小鼠诱导的抗肿瘤免疫应答。方法 将表达人成纤维激活蛋白的重组质粒CpVR-FAP混合酵母聚糖,肌肉注射免疫BALB/c小鼠3次,每次质粒100μg,酵母聚糖2mg,间隔2周。末次免疫后2周皮下接种2*104小鼠乳腺癌细胞,观察肿瘤生长体积并绘制肿瘤生长曲线,接种22天后取脾通过Elispot检测IFN-γ分泌,同时检测了脾细胞增殖情况。结果ELISPOT检测呈阳性,疫苗联合酵母聚糖免疫小鼠特异性的产生了IFN-γ(p<0.05))。在FAP抗原肽的刺激下,免疫小鼠脾淋巴细胞增殖提高10%。疫苗联合佐剂具有一定抑制肿瘤生长的能力。结论 基因疫苗 CpVR-FAP联合酵母聚糖在BALB/c荷瘤小鼠体内可诱导特异性细胞免疫免疫应答并具有抗肿瘤能力,为预防、治疗性肿瘤基因疫苗的研制提供了一定的实验依据。

Specific cellular response in tumor bearing mice induced by FAP DNA immunization combined with zymosan
Abstract:Objective: To observe the anti-tumor immune responses elicited by FAP DNA vaccine immunization with zymosan in tumor-bearing BALB/c mice. Methods: BALB/c mice were inoculated intramuscularly with the vaccine plasmid encoding FAP with zymosan, at the dosage of 100μg plasmid and 2mg of zymosan in quadriceps muscle three times at two weeks interva1.Two weeks after the last injection,2 × 104 4T1 tumor cells was injected subcutaneously into back of mice Tumor growth was measured every two days with a caliper, and the tumor volume was calculated until 22 days. After sacrifice, the spleens were collected for assays of IFN-γ secretion and the proliferation activity. Results: Enhanced specific IFN-γresponse was detected through ELISPOT in DNA+zymosan immunized mice(P<0.05) and DNA combined with zymosan improved lymphocyte proliferation by 10%. This strategy inhibited the tumor growth significantly (P<0.05). Conclusions: The DNA vaccine CpVR-FAP combined with zymosan can induce in vivo the cell mediated specific immune responses and anti-tumor effects in tumor-bearing mice, which lays a certain foundation for development of prophylactic and therapeutic tumor gene vaccines.
