ADH2基因敲除与GPD1基因沉默研究

摘要:为了高效的抑制GPD1基因的转录,在其反义序列前后加上了PGK强启动子、CYC1终止子以及KanMX 筛选标记,通过重叠延伸PCR的方法构建敲除沉默组件ADH2-PGCK,以达到在特定的区域内进行干扰和表达,而不影响其他基因的转录和表达。通过序列的分析,我们已经成功地建立了酿酒酵母ADH2敲除沉默组件,进一步将转入到酿酒酵母细胞中,并研究其表达奠定基础。

Optimize the Complex Enzymatic Preparation Silencing the Alcohol Dehydrogenase and the Glycerol-3-Phosphate Dehydrogenase Genes in Saccharomyces cerevisiae
Abstract:In order to efficiently suppress the transcription of GPD1 antisense sequence in which combined with a strong PGK promoter,CYC1 terminator and KanMX selection marker constructing ADH2-PGCK components by homologous recombination,resulting in interfering expression,without affecting other genes. By analyzing the sequences,we had successfully established ADH2-PGCK components.These findings further the development of industrial S.cerevisiae strains with improved ethanol production and reduced glycerol content for the efficient production of bio-ethanol.
