环黄芪醇对缺氧缺血后大鼠神经干细胞增殖的影响

摘要:目的 探讨环黄芪醇对神经干细胞端粒酶的激活作用,及在缺氧缺血条件下对神经干细胞增殖能力的影响。方法 传代培养神经干细胞,对细胞进行氧糖剥夺(OGD) 处理以模拟体内缺氧缺血损伤。环黄芪醇处理细胞,检测细胞端粒酶逆转录酶(TERT)表达水平及端粒酶活性;并通过检测神经球直径、Edu染色、细胞计数评价神经干细胞增殖情况。结果 环黄芪醇作用后,神经干细胞TERT表达升高,细胞端粒酶活性增加,细胞增殖能力增强。结论 环黄芪醇能激活神经干细胞端粒酶活性,促进缺氧缺血状态下神经干细胞的增殖,提示端粒酶激活剂可望用于缺氧缺血后脑损伤的修复治疗。

The effect of cycloastragenol on proliferation of neural stem cells after hypoxic-ischemic insult
Abstract:Objective In this paper,we aim to investigate the effect of cycloastragenol on telomerase activation of neural stem cells,and the proliferation of neural stem cells after hypoxic-ischemic insult. Methods The neural stem cells were cultured in oxygen-glucose deprivation (OGD) condition to imitate hypoxic-ischemic injury in vivo. They were treated with cycloastragenol (CAG, 25μM, 10μM). Then TERT expression and telomerase activity in neural stem cells were detected. Cell proliferation was examined by cell number, neural sphere diameter, and Edu staining. Results CAG increased TERT expression and telomerase activity of neural stem cells. Cell proliferation was enhanced in CAG treated neural stem cells after OGD insult. Conclusions Telomerase activator cycloastragenol increased the proliferation of neural stem cells under hypoxic-ischemic circumstances, suggesting telomerase activators might be potential agents for the therapy of hypoxic-ischemic brain injury.
