小分子化合物O-562抑制胃癌细胞增殖及机制

摘要:目的 研究小分子化合物O-562对人胃癌细胞系BGC823、SGC7901及MKN-45增殖的抑制作用及其具体机制。方法 应用不同浓度的O-562化合物处理人胃癌细胞BGC823、SGC7901和MKN-45,采用MTT法检测该化合物对细胞增殖的影响,流式细胞术检测其对细胞周期的影响,Western Blot检测该化合物对周期调控蛋白Cyclin D1和CDK4表达量的影响。结果 化合物O-562能够以剂量依赖的方式抑制人胃癌细胞BGC823、SGC7901及MKN-45的增殖,并且能够抑制上述细胞周期G1期向S期的转化。Western Blot显示O-562可以剂量依赖的方式下调胃癌细胞BGC823中Cyclin D1和CDK4的蛋白表达水平。结论 小分子化合物O-562能够通过下调胃癌细胞中Cyclin D1和CDK4的表达水平,抑制胃癌细胞G1期向S期的转化,进而抑制胃癌细胞增殖。

The effect and mechanism of O-562 suppressing the proliferation of gastric cancer cells
Abstract:Objective To investigate the effect and mechanism of O-562 on the proliferation of human gastric cancer cells. Methods MTT was used to detect the effect of O-562 on the proliferation of BGC823, SGC7901 and MKN-45 cells. Flow cytometry was used to observe the effect of O-562 on cell cycle. Western Blotting assay was adopted to check the effect of O-562 on the expression of Cyclin D1 and CDK4. Results O-562 could suppress the proliferation of BGC823, SGC7901 and MKN-45 cells in a dose-dependent manner. It could also obviously inhibit the transition of cells mentioned above from G1 to S phase. The western blot results showed that O-562 significantly down-regulated the expression levels of Cyclin D1 and CDK4 in BGC823 cells. Conclusion O-562 suppresses Cyclin D1 and CDK4 expression, in turn inhibiting the transition of cells from G1 phase to S phase, and results in the anti-proliferative effect on human gastric cancer cells.
