摘要:毕赤酵母细胞表面展示来源于鼠伤寒沙门氏菌LT2的金结合金属蛋白GolB可用于富集贵金属。本研究首次通过FACS分析和免疫荧光显微术证实目标蛋白GolB展示在毕赤酵母的表面上。然后用ICP-AES确定了重组毕赤酵母对贵金属离子的吸附能力,并通过透射电镜证实了金属离子吸附在巴斯德毕赤酵母细胞的表面上。更进一步证实展示GolB的毕赤酵母对重金属离子有一定的抗性,这可能是对贵金属离子生物吸附的一种宝贵工具。

Pichia surface display of gold binding protein for precious metal ions bio-adsorption
Abstract:A gold-specific binding metalloprotein GolB from Salmonella typhimurium LT2 was displayed on the surface of Pichia pastoris cells for enriching the precious metals. The displayed GolB was confirmed firstly through FACS analysis and immunofluorescence microscopy. Then the bio-adsorption of precious metal ions by GolB-displayed P. pastoris were determined by ICP-AES. The condition of adsorbed metal ions on the cell surface of P. pastoris was confirmed through TEM. The P. pastoris harboring cell surface displayed GolB was further shown the resistance to toxicity of heavy metal ions and it could be a valuable tool for precious metals bio-adsorption.
