柠檬醛在日本鳗鲡体内的药代动力学

摘要:本实验采用超高效液相色谱(UPLC)法研究了温度25℃-27℃、pH 6.8-7.0,以3.556 mg/L浓度的柠檬醛药浴日本鳗鲡,在不同时间段测定其体内各组织中柠檬醛药物残留量,利用DAS软件分析其消除规律。结果表明,柠檬醛在日本鳗鲡鳃、肌肉、肝脏、血浆中房室模型均为一室模型,肾脏中为二室模型。柠檬醛在日本鳗鲡鳃、血浆、肾脏、肌肉和肝脏中的药动学参数如下:达峰浓度(Cmax)分别为0.004mg/L、0.049mg/L、0.009mg/L、0.014mg/L、0.011mg/L,达峰时间(Tmax)分别为6h、6h、3h、24h、48h;表观分布容积(Vz/F)分别为3.342 L/Kg 、213.294L/Kg、1.093L/Kg、0.561L/Kg、1.191L/Kg;药时曲线下面积(AUC0~∞)分别为0.404mg/L*h、6.750mg/L*h、7.148mg/L*h、1.924mg/L*h、3.454mg/L*h;平均滞留时间(MRT0~∞)分别为196.608h、199.928h、 1200.69h、150.596h、554.578h;消除半衰期(t1/2z)分别为131.749h、140.295h、761.328h、105.131h、400.889h。药代动力学参数表明柠檬醛在日本鳗鲡体内的代谢一部分经鳃直接排出,另一部分通过血浆运送至肌肉、肝脏等其它组织,再转至肝脏并蓄积,代谢产物经肾脏排泄。

Pharmacokinetic Study of Citral in Anguilla japonica
Abstract:Applied a UPLC method to determine Citral residue in Anguilla japonica (exposed to Citral at a concentration of 4μL/L (25-27℃, pH 6.8-7.0)) every few hours and analyzed the distribution and elimination regular of Citral. The results processed by DAS software showed that the concentration-time data of Citral were described by one- compartment model in the gill, the plasma, the muscle and the liver and two- compartment model in the kidney. The main pharmacokinetic parameters in the gill, the plasma, the kidney , the muscle and the liver were obtained as following: Cmax 0.004mg/L, 0.049mg/L, 0.009mg/L, 0.014mg/L and 0.011mg/L; Tmax 6h, 6h, 3h, 24h and 48h;Vz/F 3.342 L/Kg, 213.294L/Kg, 1.093L/Kg, 0.561L/Kg and 1.191L/Kg; AUC0~∞ 0.404mg/L*h, 6.750mg/L*h, 7.148mg/L*h, 1.924mg/L*h and 3.454mg/L*h; MRT0~∞ 196.608h, 199.928h, 1200.69h, 150.596h and 554.578h; t1/2z 131.749h, 140.295h, 761.328h, 105.131h and 400.889h. The pharmacokinetic parameters showed that the metabolic pathways of Citral in Anguilla japonica was that some were absorbed by the gill, another were transported to different tissues(include the muscle) and cumulated by the plasma, then move into the liver, finally excrete by the kidney.
