生、蜜百部总生物碱对哮喘小鼠免疫作用差异研究

摘要:目的:本文采用OVA 激发哮喘小鼠模型研究生、蜜百部总生物碱抗免疫作用药理学的差异性,通过液质联用技术测定其生物碱成分变化,探讨其产生药理变化可能的物质基础。方法:口服给予不同浓度生、蜜百部生物碱后测定哮喘小鼠脾细胞中Th1 及Th2 释放的细胞因子,采用RT-PCR 技术测定实验小鼠肺组织中GATA-3 及T-bet 基因表达。结果:模型组与空白组比较显著降低了IFN-γ、IL-2及IL-13含量,升高了IL-4及IL-5含量(P < 0. 05);与生品比较,炙品总生物碱中、高剂量组给药后更能上调哮喘小鼠脾细胞中炎症因子IFN-γ( P < 0. 05) ,高、中、低剂量炙品总生物碱组显著下调IL-5( P< 0. 05) ;PCR实验结果显示炙碱给药组更能上调T-bet /GATA-3( P < 0. 05) 表达。结论:百部总生物碱提取物可通过调节Th1 /Th2 比例治疗哮喘,但制品作用更为显著,可能是因为百部炮制后主要生物碱类成分比例发生变化引起的,为进一步揭示百部炮制原理奠定基础。

Immunization difference of total alkaloids extracted from original and processed Stemona Tuberosa on asthma rat
Abstract:Immunization difference of total alkaloids extracted from crude and processed Stemona tuberose was studied based on OVA-induced asthma mice model, and chemical difference was analyzed by UPLC-MS to explore the material foundation for pharmacological change. Mice were orally administered with different concentration of total alkaloids, and the cytokine levels from T-cells in splenocytes were determined by ELISA. RT-PCR was carried out to describe the ratio of T-bet /GATA-3. The results showed a significant decrease in IFN-γ, IL-2, IL-13 levels and an increase in IL-4 and IL-5 levels in the model group. Compared with crude extract, middle and high group of processed samples significantly increased the levels of INF-? (P<0.05)and all three groups decreased that of IL-5(P<0.05). The PCR test results showed processed samples increased the ratio of T-bet /GATA-3(P<0.05). This study indicated that total alkaloids extract from crude and processed Stemona Tuberosa could treat asthma by regulating Th2/Th1 imbalance and the processed one had a more significant effect, which might be caused by the change in the ratio of alkaloids after being processed This paper lays the foundation for further revealing the processing principle of Stemona Tuberosa and provides the experimental support for further development of antitussive medicine of Stemona Tuberosa.
