赖氨酸甲基转移酶Kmt5a在猪睾丸的表达模式及剪接变体结构分析

摘要:赖氨酸甲基转移酶KMT5a参与组蛋白甲基化修饰、细胞分裂、细胞凋亡等多种生理活动,为了揭示Kmt5a基因在猪睾丸中的表达模式,本研究克隆获得了猪睾丸Kmt5a基因的不同转录本Kmt5a-X1、Kmt5a-X2序列,检测了Kmt5a在猪睾丸发育过程中相对表达量的变化,分析了剪接变体间的结构差异。结果表明,Kmt5a在7日龄猪睾丸支持细胞(Sertoli cells)表达量显著高于间质混合细胞(Interstitial cells)和性原细胞(Gonocytes),在5月龄猪睾丸中相对含量显著高于7日龄与2月龄。蛋白二维结构分析表明KMT5A-X1缺少N端的PIP (PCNA- interacting motifs) box1,并且N端的Cdk1/Cyc B 结合序列处在第二位氨基酸位置,推测KMT5A-X1与Cdk1/Cyc B的结合受到影响。

The Expression Pattern and Splicing Variant Analysis of Lysine Methyltransferase Kmt5a in Pig Testicles
Abstract:Lysine methyltransferase 5A (KMT5A) plays an important function in histone methylation, cell division, apoptosis and other physiological activities. This study interpreted the expression pattern of Kmt5a gene in pig testicles by cloning two different splicing variants Kmt5a-X1 and Kmt5a-X2, detecting the relative expression quantity change of Kmt5a gene in the development process of pig testicles, analysing of the structural differences between the splicing variants. The results showed that the relative expression quantity of Kmt5a in 7-day-old sertoli cells were significantly higher than which in interstitial cells and gonocytes of testicles. The global level of Kmt5a relative expression quantity in 5-month-old testicles was significantly higher than those in 7-day-old and 2-month-old. The analysis showed that protein KMT5A-X1 was lack of PIP (PCNA-interacting motifs) box1 in the N-terminal compared with KMT5A-X2. The serine, a potential phosphorylation sites of Cdk1/Cyc B complex, was located at amino acid residues 2 of KMT5A-X1. Presumably its special position could affect the ability to combine with Cdk1/Cyc B complex.
