芝麻素改善压力负荷诱导的心肌肥厚

摘要:目的:本研究旨在探讨芝麻素对心肌肥厚小鼠模型的影响及可能的机制。方法:主动脉缩窄术构建小鼠心肌肥厚模型,分为假手术组,假手术+芝麻素治疗组,主动脉缩窄组,主动脉缩窄+芝麻素治疗组。术前3天给予芝麻素口服。超声心动图和血流动力学检测评价芝麻素对大鼠心脏收缩及舒张功能的影响;通过Western blot检测心肌组织中分子水平表达变化;通过RT-PCR检测心肌组织中转录水平的表达变化;HE染色和PSR染色观察形态学变化。结果:主动脉缩窄4周后,芝麻素治疗组的MEK和ERK蛋白表达显著低于假手术组和主动脉缩窄组。给予芝麻素治疗后心肌肥厚程度减轻,心肌肥厚及纤维化相关标志物的转录水平较未给予治疗的主动脉缩窄组降低;PSR染色胶原提示治疗后胶原生成减少。结论:芝麻素减轻心肌肥厚以及心肌纤维化改善心脏功能。在压力负荷诱导的心肌肥厚中,芝麻素可能通过抑制ERK信号通路发挥对心脏的保护作用。

Sesamin attenuates pressure overload-induced cardiac hypertrophy
Abstract:The aim of this study was to investigate the effects of sesamin on the remodeling of myocardium in mice and the possible mechanisms. Transverseaortic constriction (TAC) was performed to induce cardiac hypertrophy in mice, and sesamin was administered for 4 weeks. Mice were allocated into four groups: Sham + vehicle, TAC + vehicle, sham + sesamin and TAC + sesamin. Sesamin treatment attenuated cardiac dysfunction, as indicated by the results of echocardiography and catheter based measurements at 4 weeks post surgery. Sesamin inhibited the expression of collagen Iα and collagen III and interstitial fbrosis. In addition sesamin downregulated the activation of the extracellular signal regulated kinase (ERK), In conclusion, sesamin attenuated cardiac hypertrophy and interstitial fbrosis, as well as improving left ventricular function in pressure-overloaded mice.
