丁香抑菌活性及机理的初步研究

摘要:丁香提取物以其较好抗菌功效而具有广阔的医用前景,但目前尚未有较深入的抑真菌机理类研究报道.本研究以真菌代表菌属酿酒酵母为例,于分子水平阐释了丁香对菌体的抑菌机理。结果表明,丁香对所试菌种均有较好的抑菌活性,PI染色分析与电镜扫描结果显示丁香提取物对细胞壁与细胞膜均有损坏作用,同时蛋白凝胶电泳结果表明真核翻译延长因子eEF1A蛋白表达减弱。而其相应的调控基因TEF1与TEF2在RNA水平无显著性变化。综上所述,丁香提取物在蛋白水平上抑制了eEF1A的表达,而没有对其调控基因在转录水平产生影响。抑制eEF1A蛋白的表达可能是丁香抑制酿酒酵母生长的一部分原因。本研究为丁香提取物作为天然抗菌剂应用于医药领域提供了新的依据。

Antifungal activity of clove extract and Its Mechanism of Action
Abstract:Clove extract has great application potential as a natural antibiotics agent. However, there are few reports about the mechanisms of antifungal activity of clove extracts. We used S. cerevisiae as a fungal model to address the molecular process of the antifungal activity exerted by clove extract in current study.The results showed clove extract had significant effects on all of the tested strains. Propidium iodide staining and Scaning electron micrographassays showed the damages of cell wall and membrane in S. cerevisiae by the addition of clove extract. Furthermore, SDS-polyacrylamide gel electrophoresis assays revealed that the protein expression of eukaryotic elongation factor 1 alpha (eEF1A) was suppressed significantly after treatment with clove extract, while its corresponding gene had no significant change. These results showed that clove extract plays a role by down-regulating yeasteEF1A protein expression level, but not at the transcriptional level. The down-expression of eEF1A protein may contribute to its antifungal activity. These results may have clinical significance for future application of clove extract as a natural antifungal agent.
