脂肪酸合酶抑制剂的筛选及其对LNCaP细胞增殖的抑制作用

摘要:脂肪酸合成酶(FASN)是人体内脂肪酸合成的关键酶,同时也是治疗癌症的潜在靶点。本研究通过实验室开发的一种微板筛选法快速地从药食同源食物中筛选到了一种脂肪酸合成酶抑制剂-丁香乙醇提取物(AEC)。实验表明该提取物能够降低高山被孢霉及人前列腺癌细胞LNCaP中棕榈酸(PA)的含量。进一步探究AEC对FASN的抑制作用,发现AEC能够下调LNCaP肿瘤细胞FASN蛋白的表达水平,干扰肿瘤细胞的细胞周期及抑制其增殖。丁香乙醇提取物作为一种新型的天然FASN抑制剂,具有抑制内源性脂肪酸合成和抑制肿瘤细胞增殖的作用。

Fatty acid synthase inhibitor screening and its inhibitory effect on LNCaP cell proliferation
Abstract:Fatty acid synthase (FASN) is the key enzyme of de novo lipogenesis and a potential cancer therapeutic target. Using the fast microplate screen method developed in our laboratory, from various natural edible plants, we discovered that alcohol extract of clove (AEC) has the inhibitoryeffecton FASN expression . In this study, AEC treatment could reduce the content of palmitic acid (PA) both in M. alpina and in the human prostate cancer cell. Moreover, AEC not only inhibited the FASN expression in LNCaP cell, but also suppressed the proliferation and cell cycle in LNCaP cells. Thus AEC may be a potential natural cancer therapeutic agent as a FASN inhibitor.
