枯草芽孢杆菌产脂肽的发酵优化

摘要:近年来,脂肽作为一种新型的生物表面枯草芽孢杆菌产脂肽的发酵优化枯草芽孢杆菌产脂肽的发酵优化枯草芽孢杆菌产脂肽的发酵优化活性剂,具有抗菌、抗癌、抗病毒及溶血等作用,正逐渐被应用于食品加工与保鲜、农业生物防治、原油开采及医药等领域,但是较低的产量无法满足工业化的需求。本实验利用较低成本的底物对枯草芽孢杆菌ATCC21332进行发酵优化,从而得到枯草芽孢杆菌在葡萄糖含量在9g/l,脱脂豆粕含量在40g/l,接种量在5%,装液量为120ml(500ml的容量瓶),温度为30 C,转速为150r/min,发酵时间为120h时脂肽产量最高为4885mg/l,相比Yeh通过添加活性炭载体达到的最高产量3600mg/l增加了35.70%。同时通过测定发酵过程中不同时间段的发酵液中氨基酸的含量,发现在86h时谷氨酸含量急剧增大,同时发酵液表面张力由72下降到28 mN/m,并且通过定量添加氨基酸,发现添加谷氨酸可使脂肽产生量达到954mg/l,相比不添加谷氨酸的培养基可使脂肽产生量增加6倍。从而为工业化生产提供新思路。

Enhanced production of surfactin by Bacillus subtilis ATCC 21332
Abstract:Biosurfactants, an alternative of chemically synthesized surfactants, has been widely used in many commercial areas such as petroleum, pharmaceuticals, biomedical, and food processing industries. This study investigate that alternative medium options are being sought to improve the production of surfactin with relatively accessible and low-cost raw materials. It turns out that the production of surfactin was up to 5483.2mg/l under the optimization condition that the soybean meal as the substrate of 40g/l, glucose concentration of 7g/l, incubation volume of 5%, the dissolved oxygen content of 100ml (per 500ml flask), which increased by 52.3% enhanced production of surfactin by Bacillus subtilisATCC 21332 when compared with the yield of surfactin 3600mg/l before. In addition, by addition of the specific amino acids to the medium, the yield of surfactin was increased by different extent, among of which is the glutamate that have the maximum yield of surfactin up to 954.395mg/l, which is 7 times content of the control. What's more, the antibacterial properties of surafctin and the ability that can decrease the surface tension of water from 72 to 28 mN/m also give us an insight of the prospect that surfactin can be used in the area of food and other fields.
