金钗石斛水提物体外降胆固醇作用研究

摘要:体外模拟人体肠道环境,研究了金钗石斛水提物对胰脂肪酶活性、胆固醇酯酶活性和胆固醇胶束溶解度的影响及其与胆酸盐的结合能力。结果表明:金钗石斛水提物对胰脂肪酶和胆固醇酯酶的活性都有抑制作用,抑制效果在一定浓度范围内呈剂量依赖性;10 mg/mL的金钗石斛水提物与牛磺胆酸钠、甘氨胆酸钠和胆酸钠都有一定的结合能力,结合率分别为50.90%、76.08%、76.65%;金钗石斛水提物对胆固醇胶束化溶液有较强的结合能力,可以降低胆固醇胶束的溶解度,其半数抑制浓度IC50为17.08 mg/mL。结论:金钗石斛水提物可以抑制胰脂肪酶和胆固醇酯酶活性并结合胆固醇胶束和胆酸盐,减少人体对胆固醇的利用和吸收,从而起到降胆固醇的作用。

Cholesterol-Lowering Effect of Dendrobium nobile Aqueous Extract in Vitro
Abstract:The study investigated the effect of Dendrobium nobile aqueous extract on the inhibition of pancreatic lipase and cholesteroL esterase activty and their binding capacity to bile acid and cholesterol micellization. The results indicated that Dendrobium nobile aqueous extract inhibited the activity of pancreatic lipase and cholesteroL esterase. At the concentration of 10 mg/mL, the binding rates of Dendrobium nobile aqueous extract to sodium taurocholate, natrium glycocholicum and sodium cholate were 50.90%, 76.08%, 76.65% respectively. Dendrobium nobile aqueous extract also inhibit cholesterol micellization formation effectively and the 50% inhibiting concention(IC50, mg/mL)was 17.08mg/mL. It was suggested that Dendrobium nobile aqueous extract may decrease lipid obsortion by inhibiting the activity of pancreatic lipase and cholesteroL esterase, and bingding bile acid and cholesterol micellization, and therefore may apply for their cholesterol-lowering effect in vivo.
