生物纳米分辨率动态三维相衬成像技术

摘要:为了实现疾病的早发现早治疗,人们迫切需要从分子细胞水平去认识生命的本质,探求疾病的发生演化机理。相衬成像技术可以应对"在维持细胞活性的前提下,实时、准确地检测活体细胞内部分子的三维空间位置动态分布"的挑战。目前,相衬成像正在进一步向广度和深度发展,有望在以下三个亟待解决的关键科学问题方面取得重大突破:3D相衬成像理论、非相干光源3D相衬成像、动态3D相干衍射成像。将高亮高相干超快X光源技术、相衬成像技术、微纳光学技术和3D重建成像技术相结合,催生了纳米空间分辨3D动态相衬成像技术的快速发展。这类前沿的相衬显微技术,将突破光学显微镜的分辨率低和电子显微镜穿透性差的限制,有望实现活细胞的"动态、3D、纳米空间分辨率成像观测"这一人们期待以久的梦想,为生命科学、材料科学、信息科学、能源科学和纳米科技等关键学科领域提供最先进的研究手段。

Dynamical Three-dimensional Phase-contrast Nano-imaging for biopsy
Abstract:In order to achieve early detection and treatment of diseases, there is an urgent need to explore the mechanism of diseases occurrence and evolution and to understand the essence of life on the molecular and cellular level. The challenge we are facing is to invasively detect the accurate three-dimensional position of molecules within living cells in real-time. In order to overcome this problem, scientists are exploring phase-contrast microscopy for dynamical three-dimensional nano-imaging of living cells. This new 3D nano-imaging will break through the current limitations of microscopy confused by in vivo observation of living cells and provide important theoretical supports and reliable solutions for cancer biopsy and early diagnosis of diseases.
