干扰脑微血管内皮细胞中ATG7的表达导致细胞迁移能力下降的机制研究

摘要:(目的研究自噬相关基因ATG7对脑微血管内皮细胞迁移、增殖能力的影响及调控机制。方法构建稳定敲降ATG7的脑微血管内皮细胞,利用WST实验、划痕实验、EMSA检测ATG7对脑微血管内皮细胞迁移、增殖能力的影响。结果ATG7表达下降不影响HBMEC的增殖能力;ATG7表达下降导致HBMEC的迁移能力下调。结论脑微血管内皮细胞中自噬相关基因ATG7可能通过NF-κB信号调节细胞的迁移能力。

Interfering ATG7 down-regulates the migration of HBMEC
Abstract:In this paper, we constructed a stable ATG7 knock-down human brain microvesicular endothelial cell (HBMEC) via shRNA. Using this cell, we performed the proliferation and migration study of ATG7 on HBMEC. The results showed that interfering ATG7 could significantly reduce the migration ability of HBMEC but not the proliferation ability.The migration of HBMEC was ATG7-mediated NF-κB-Dependent manner.
