蛋白质多态解折叠行为与稳定性,III解折叠机理与物种分布

摘要:基于结构基元模型,由n个不同结构基元组成的蛋白质或酶,化学变性剂诱导的解折叠曲线表现为(n+1)态,其解折叠过程可按照"连续反应"和"平行反应"两种机理进行,"连续反应"机理的(n+1)态有(n+1)种物种,而"平行反应"机理的(n+1)态有2n种物种;推导出两种反应机理下蛋白质或酶解折叠时物种分布随化学变性剂浓度变化的表达式;以尿素、盐酸胍诱导的卵清溶菌酶解折叠为例,用光谱、酶活性测定的解折叠曲线分析了两种化学变性剂诱导溶菌酶解折叠的不同反应机理;蛋白质或酶结构基元的[D]1/2、化学变性剂等是影响蛋白质或酶解折叠反应机理的重要因素。

Stability of Proteins with Multi-state Unfolding Behavior, IIIThe Mechanism of Protein Unfolding and Species Distribution
Abstract:Based on the model of structural element, When a protein consists of n different structural elements its unfolding will shows a (n+1)-state behavior. The unfolding mechanisms include sequential and parallel mechanisms. According to sequential mechanism, if the protein shows (n+1)-state, it will contain n+1 species. However, in accordance with parallel mechanism, (n+1)-state shows 2n species. The specises distrbution of protein or enzyme under different denaturation concentration in two unfolding mechanisms is deduced. Taking the unfolding of hen egg white lysozyme induced by guanidine hydrochloride and urea as an example, the two different unfolding mechanisms of hen egg white lysozyme are analyzed using spectra and enzyme activity methods. The protein or enzyme unfolding mechanisms are deeply affected by the [D]1/2 of protein or enzyme structural element and chemical denaturation.
