川芎根腐病拮抗性内生放线菌FL9活性代谢产物初探

摘要:目的:对川芎根腐病拮抗菌株(FL9)次生代谢产物的生物活性及结构类型进行探究。方法:采用纸片法对FL9摇瓶发酵产物生物活性及提取分离条件进行考查;再通过扩大培养制备大量发酵液,结合硅胶柱层析、HPLC等方法分离纯化代谢产物;最后对产物进行活性验证及LC-MS检测分析。结果:FL9的次生代谢产物具有抗根腐病病原菌的活性;从FL9代谢产物中初步分离获得H5-1及H5-2两个成分。结论:结合文献及图谱分析,二者基本结构由羟基蒽环和糖基组成,属于蒽环类抗生素或其衍生物。

Preliminary study on the metabolites of Antagonistic Endophytic Actinomycetes FL9 to root rot of Ligusticum chuanxiong Hort.
Abstract:The purpose of this study was to investigate the biological activity and structure types of the secondary metabolites of antagonistic bacteria (FL9) isolated from Root Rot of Ligusticum chuanxiong hort.. the paper method was used to detect and track the biological active substances. The purity was determined by high performance liquid chromatography (HPLC) with thin layer chromatography and column chromatography. The active ingredients were prepared by preparative chromatography, and the pure substances were obtained by crystallization and recrystallization. Finally LC-MS (measured with molecular weight of 620 and 516). Based on the analysis and comparison of the structures of the compounds, H5-1 was confirmed to be molecular C41H36N2O4 and H5-2 as molecular C25H32N4O8.
