丹参酚酸B上调BCG感染巨噬细胞激活性NKR配体以促进NK细胞活性

摘要:目的探讨结核分枝杆菌感染对人巨噬细胞激活性NK细胞受体(NKR)配体表达及诱导NK细胞活化效应的影响;以及丹参酚酸B对感染细胞相应配体表达及NK细胞杀伤活性的潜在干预效应。方法诱导THP-1细胞分化为巨噬细胞,建立BCG感染的人巨噬细胞模型,并用丹参酚酸B体外干预感染细胞,使用Western blotting和Real-time PCR的方法分别检测巨噬细胞上ULBP1、ULBP2、MICA和MICB蛋白和mRNA水平。以BCG感染THP-1为靶细胞,人PBMC诱导的NK细胞为效应细胞,共孵育后,RTCA DP实时无标记细胞分析仪检测NK细胞的杀伤活性。结果BCG可促进巨噬细胞ULBP1、ULBP2、MICA和MICB的mRNA水平上调,并上调MICA和ULBP1的蛋白表达。1mg/L丹参酚酸B干预BCG感染巨噬细胞72h后,可显著上调ULBP1和MICA的蛋白和mRNA表达水平,并增加NK细胞对靶细胞的杀伤率。结论结核分枝杆菌感染可上调人巨噬细胞激活性NKR配体的表达,但没有促进NK细胞活化;丹参酚酸B具有促进上调感染细胞激活性NKR配体表达及NK细胞杀伤活性的干预效应。

Salvianolic acid B regulates NKR ligands on BCG infected Macrophages and promotes NK activity
Abstract: Objective To study the effect of Mycobacterium tuberculosis (Mtb) on the expression of Ligands of activating NK receptors (NKRs) on human macrophages infected with Mtb and the activation of NK cells, and the potential effect of Salvianolic acid B on Ligands expression and NK cell activity. Methods THP-1 derived macrophages were induced and infected with BCG and treated with Salvianolic acid B, then the protein and mRNA level of ULBP1, ULBP2, MICA and MICB were evaluated by using Western blotting and Real-time PCR. BCG infected macrophages were as target cells, and NK cells derived from human PBMC were as effective cells, and then measured the killing rate of NK cells by RTCA DP Real-time Cellular Analysis after co-cultured with target cells. Results BCG infection up-regulated the mRNA levels of ULBP1, ULBP2, MICA and MICA on macrophages, furthermore promoted the protein expression of MICA and ULBP1. Salvianolic acid B (1mg/L) increased the mRNA and protein level of ULBP1 and MICA and the killing rate of NK cells 72h after treatment remarkably. Conclusion Ligands of KARs on human macrophage but the NK activity could be up-regulated by BCG infection, Salvianolic acid B could promote the killing activity of NK cells by increasing the expression of ligands of KARs on BCG infected cells.
