UVB对绵羊黑色素细胞中GPNMB和PMEL的影响

摘要:非转移性黑色素瘤糖蛋白b(Glycoprotein nonmetastatic melanoma protein b,GPNMB)及其同系物色素细胞特异性黑色素细胞蛋白(Pigment cell-specific melanocyte protein,PMEL)对黑素体的形成和黑色素的生成具有重要的作用。本文旨在探索GPNMB和PMEL在不同毛色绵羊皮肤是否存在差异表达,确定两者与黑色素生成的关系以及两者之间是否存在相关性。免疫组织化学结果表明GPNMB在不同毛色绵羊皮肤的毛基质、内外毛根鞘、毛乳头等区域均有表达。Western blot和QRT-PCR结果显示黑色绵羊皮肤中GPNMB和PMEL表达量高于白色绵羊皮肤。为了进一步了解GPNMB和PMEL对黑色素沉着的影响以及两者之间的关系,我们通过UVB对黑色素细胞处理,同时检测GPNMB和PMEL的表达量变化。结果表明UVB在100 mJ/cm2剂量时黑色素细胞活性最高;UVB辐射一次后GPNMB和PMEL mRNA表达量在72 h达到顶峰,并且两者的曲线变化趋势大致相同。以及UVB连续辐射GPNMB mRNA表达量在4d达到顶峰,而PMEL mRNA表达量在3 d最高,并且与GPNMB和PMEL的曲线变化不一致。由此表明GPNMB和PMEL在不同毛色绵羊皮肤存在差异性表达,UVB对GPNMB和PMEL产生不同的影响,GPNMB和PMEL可能通过某种通路影响黑素体的成熟来调节其和黑色素的生成,进而参与毛色的形成。

Effects of UVB on GPNMB and PMEL in Sheep Melanocytes
Abstract:Glycoprotein nonmetastatic melanoma protein b(GPNMB) and pigment cell-specific melanocyte protein(PMEL) play an important role in the formation of melanosomes and the melanogenesis.To investigate whether GPNMB and PMEL differentially expresses in the skin of sheep with various coat colors and to determine the relationship between GPNMB and PMEL generation with melanin and whether there is a correlation between GPNMB and PMEL.Immunohistochemical assays revealed that GPNMB was expressed in the hair follicle matrix, the inner and outer root sheath, and the dermal papilla in the skin with various coat colors.The results of Western blot and QRT-PCR analysis showed that the expression of GPNMB and PMEL in black sheep skin was higher than that in white sheep skin.In order to further understand the impact of GPNMB and PMEL on melanin pigmentation and the relationship between two.We treated the melanocytes by UVB and detected the expression levels of GPNMB and PMEL.The results showed that the activity of melanocytes was the highest at 100 mJ/cm2 dose,the expression of GPNMB and PMEL mRNA reached the peak at 72 h after UVB irradiation, and the curve trends of GPNMB and PMEL are roughly the same. The expression of GPNMB mRNA in UVB continuous radiation reached its peak at 4 days, however, the expression level of PMEL mRNA was the highest at 3 d, and it was inconsistent with the curve of GPNMB and PMEL.The results showed that GPNMB and PMEL were differentially expressed in different coat sheep, UVB has different effects on GPNMB and PMEL,GPNMB and PMEL were involved in the formation of coat color by regulating the maturation of melanosomes through a pathway to regulate the formation of melanin.
