摘要:肿瘤报告探针有一种酸响应信号触发的高分子探针在生物成像中的应用一种酸响应信号触发的高分子探针在生物成像中的应用助于指导切除手术过程中肉眼不可见的肿瘤病灶,而由于肿瘤表面存在不同的凝集素,可以作为探针的靶向目标。我们报道了一组用于小鼠肿瘤成像的含糖高分子胶束。这些胶束由对溶酶体酸度起反应的长沙红为核心和与不同半乳糖胺糖基化的聚[苯乙烯-交替-马来酸]壳层组成。这些纳米级胶束在细胞外无荧光,在酸性溶酶体内发出强烈荧光,从而可以在光学层面跟踪肿瘤对胶束的内吞作用。体内筛选发现修饰α构型半乳糖胺的高分子胶束可以被小鼠肿瘤高效摄取并激活荧光,且不易积聚在肿瘤中。鉴于该类高分子探针对肿瘤的高效靶向性,凝集素靶向信号触发型胶束对诊断或手术切除肿瘤具有一定意义。

A targetable acid-responsive micellar system for tumor imaging
Abstract:Tumor-reporting probes are valuable to guide surgical resection of tumor foci elusive to visual inspection. As tumors display distinct arrays of lectins, we herein report the construction of a panel of glycan-displaying micelles for tumor illumination in mice. These micelles consist of cores of Changsha-red responsive to lysosomal acidity and a corona of poly[styrene-alter-(maleic acid)]glycosylated with different galactosamine. These nanoscale micelles are nonfluorescent extracellularly and become luminescent within acidic lysosomes, enabling optical tracking of tumor endocytosis of the micelles. In vivo screening revealed high-efficiency uptake and fluorescence activation of galactosylate micelles by subcutaneous tumor. Given the robustness and high performance of this system, lectin-targA targetA targetable acid-responsive micellar system for tumor imagingable acid-responsive micellar system for tumor imagingeted responsive micelles are attractive for diagnosis or surgical ablation of tumors.
