鼎湖鳞伞菌丝体粗多糖的流变学特性研究

摘要:采用液体深层发酵制备鼎湖鳞伞菌丝体,再用热水浸提、乙醇沉淀的方法获得粗多糖(PDP)。在静态流变学性质测试,考察了浓度、温度、pH值、蔗糖及无机盐添加量(NaCl及CaCl2)对粗PDP溶液表观粘度的影响,结果表明粗PDP溶液的表观粘度随着多糖浓度的增加而上升,随着剪切速率的增加而快速下降,说明粗PDP溶液是假塑性流体(非牛顿流体);多糖溶液的表观粘度在pH= 6-10的范围内保持稳定,而在强酸性条件下则明显上升;添加低浓度CaCl2,粗PDP粘度变化不明显,但随着浓度的继续增加而逐渐增大。因此,粗PDP溶液具有较好的耐热、耐碱及耐盐稳定性。

Rheological property of mycelial polysaccharides from Pholiota dinghuensis Bi
Abstract:The mycelia of Pholiota dingensis Bi were produced by the submerged fermentation and crude polysaccharides (PDP) were obtained by hydrothermal extraction and ethanol precipitation. Effects of concentration, temperature, sucrose, pH, NaCl and CaCl2 on the static rheological property of crude PDP were investigated. The results showed that the apparent viscosity of crude PDP increased with the increase of crude PDP concentration, while decreased significantly with the increase of shear rate, indicating that crude PDP solution was a non-Newtonian pseudoplastic fluid. The apparent viscosity of crude PDP remained stable under pH of 6-10, while increased significantly with a strong acid treatment. The rheological property of crude PDP did not changed in a low concentration of CaCl2, and gradually increased with a higher concentration of CaCl2. Therefore, crude PDP was resistant to heat, alkali and salt.
