植物乳杆菌ZJ316黏附蛋白基因的克隆及序列分析

摘要:本研究以本实验室保存的从婴儿粪便中筛选得到的一株产细菌素的植物乳杆菌(Lactobacillus plantarum ZJ316)为研究对象。采用同源克隆法,根据Lactobacillus plantarum WCFS1表层黏附蛋白Slp2588、黏液黏附蛋白Muc2486 的基因序列(1062815)、(1062646)进行引物设计,从Lactobacillus plantarum ZJ316中克隆得到了长度为1618 bp的基因片段slp2588A和 3018 bp的基因片段muc2486A。序列分析表明,基因片段slp2588A和Lactobacillus plantarum WCFS1中的黏附基因slp2588相似性为98%;基因片段muc2486A和L. plantarum WCFS1中的黏附基因muc2486相似性为99%。

CLONING AND SEQUENCE ANALYSIS OF ADHESION PROTEIN GENE FROM L.PLANTARUM ZJ316
Abstract:In this study, Lactobacillus plantarum ZJ316 is preserved in our laboratory,which isolated from infant fecal samples and produce bacteriocin. According to Lactobacillus plantarum WCFS1, whose adhesion protein genes sequence (1062815) and (1062646) of surface adhesion protein Slp2588, mucous adhesion protein Muc2486 for primer designing, we carried on the adhesion gene cloning and sequencing. Sequencing results: The length of Gene fragment slp2588A is 1618 bp and The length of gene fragment muc2486A length is 3018 bp. Similarity comparison in the NCBI website with BLAST software, the result showed that the similarity of gene fragment slp2588A and gene slp2588 in Lactobacillus plantarum WCFS1 was 98%; Similarity comparison in the NCBI website with BLAST software, the result showed that the similarity of gene fragment muc2486A and gene muc2486 in Lactobacillus plantarum WCFS1 was 99%.
