噬菌体展示筛选与狂犬病毒P互作的宿主蛋白

摘要:狂犬病是一种人或其他温血动物的病毒性脑脊髓炎,在家畜和野生动物之间可以自然感染与传播,感染后一旦出现临床症状,致死率几乎100%。其病原体是弹状病毒科狂犬病毒属的狂犬病毒(Rabies virus, RABV)。狂犬病毒有五种结构蛋白,其中磷酸化蛋白P是一种多功能的非催化活性蛋白,在病毒的转录和复制过程中发挥重要作用,而狂犬病毒的致病机制仍未完全阐明。本研究首先表达纯化了狂犬病毒P蛋白,以纯化的P蛋白为靶蛋白,通过T7噬菌体展示的方法筛选相互作用蛋白。通过5轮的淘选,筛选得到多个阳性克隆,其中核糖体大亚基蛋白L9(RPL9)与P蛋白特异性相互作用,并通过ELISA、GST pull-down得到验证,为进一步阐明狂犬病毒的复制和致病机理奠定基础。

Screening the Host Proteins Interaction with Rabies Virus P by Phage Display System
Abstract:Rabies is a kind of viral encephalomyelitis for people or other warm-blooded animals. once appear clinical symptoms, the fatality rate is almost 100%. The pathogen is Rabies virus, which belong to the Lyssavirus genus of the Rhabdoviridae family. Rabies virus is composed of five structural proteins. The phosphorylated protein (P), which is an uncatalytic activity protein, plays an important role in the virus replication, transcription and interferon antagonism. Moreover, the pathogenesis of rabies virus is still far from understanding. To identify a cellular protein(s) that interacts with RABV P protein, we performed a phage display-based protein interaction screen. Through five rounds of elutriation, we obtained several positive clones. And the results show that RPL9 is the target of P protein. Binding between RPL9 and P was also demonstrated by ELISA, GST pull-down. Our study laid the foundation to further clarify the mechanism of rabies virus replication and pathogenesis.
