DNA甲基化在UV诱导皮肤癌变中的作用

摘要:紫外线辐射(UVR)诱导皮肤发生癌变涉及到多个基因复杂变化过程,且其中的分子机制也十分复杂。越来越多的研究表明UVR诱导皮肤癌变有DNA甲基化的参与,UV诱导基因启动子的甲基化和全基因组的甲基化水平而参与皮肤的肿瘤形成。虽然目前有报道UVR诱导DNA甲基化,但是很多结论相互矛盾,并且UV诱导DNA甲基化的机制尚不明确。本文首次系统从UV导致DNA损伤和UV对抑癌基因的调控方面介绍DNA甲基化在其中存在的作用,并讨论了UV诱导DNA甲基化可能存在的分子响应机理。以期对研究UVA导致皮肤癌变的机制研究提供新的视角和基础材料。

DNA Methylation Changes in UV Inducing Skin Carcinogenesis
Abstract:Ultraviolet radiation (UVR) inducing skin carcinogenesis is a complex process which is involved multiple genes, as well as molecular mechanism. A growing number of studies have shown that UVR induced s skin carcinogenesis with the participation of DNA methylation. UV induced gene's promoter methylation and Global methylation. Although UVR induction of DNA methylation has been reported, many conclusions is contradict each other and the mechanism of UV induced DNA methylation is unclear.This review is systemly from UV causing DNA damage and UV causing disregulation of tumor suppressor gene to introduce DNA methylation's fuction of it. And the possible molecular mechanism of UV induced DNA methylation is discussed, with a view to providing new perspectives and basic data for the academic research of UV inducing skin carcinogenesis.
