新型Cys荧光探针的设计、合成与应用研究

摘要:本工作设计了一种新型的双光子检测半胱氨酸(Cys)的荧光探针P2。该探针是通过将Cys响应基团丙烯酰基引入到双光子荧光团6-羟基-2-乙酰萘上设计而成的。在375 nm光激发下,该探针本身的荧光十分微弱,与Cys反应后,探针P2在455 nm处有一个明显的荧光增强(20 倍)。探针对Cys的高响应倍比使得探针检测限(S/N)很低,为0.10 μmol/L。此外,探针对Cys的响应迅速(28 min 内荧光强度达到饱和),在生理pH条件下表现出高稳定性、高选择性。更有意义地是,探针P2具有高选择性,低毒性和好的生物相容性,能够用于活细胞中Cys的双光子成像。

Design, synthesis and application of fluorescent probes for Cys
Abstract:In this work, a novel two-photon probe P2 has been developed for cysteine (Cys) detection. The probe was based on introducing a Cys-triggered acryloyl group to the two-photon fluorophore 1-(6-hydroxynaphthalen-2-yl) ethanone. The probe P2 itself exhibited weak fluorescence, however, it displayed a dramatic fluorescence enhancement at 455 nm responding towards Cys (about 20 fold) and the high signal to background ratio enables a low limit of detection(S/N=3) to be 0.10 μmol/L. In adittion, the probe presents a fast response which the fluorescence intensity of the probe tends to stable in 28 minutes, high stability and high selectivity towards Cys at physiological pH conditions.More meaningfully, the probe P2 could be used for two-photon imaging of Cys in living cells with high selectivity, low toxicity and good biocompatibility.
