环状RNA在肝癌中的作用及机制研究

摘要:目的:探究环状RNA在肝癌中的作用及机制。方法:通过circinteractome数据库预测与Lin28结合的circRNA;由RIP实验及qRT-PCR确定Lin28与所选基因的circRNA结合情况;由环状RNA慢病毒表达载体PLO-ciR构建环状RNA过表达载体;qRT-PCR检测肝癌细胞系中环状RNA过表达水平;Western blot检测环状RNA过表达肝癌细胞系中Lin28蛋白表达水平;结果:circRNA与RNA结合蛋白Lin28存在相互结合;成功构建circRNA过表达载体;circRNA在肝癌细胞系中有显著的过表达效果;circRNA过表达的肝癌细胞系中Lin28蛋白表达下降。结论:circRNA在肝癌细胞系中发挥调控Lin28表达的作用。

The role and mechanism of circular RNA in hepatocellular carcinoma
Abstract:Objective: To explore the function and mechanism of circular RNA in hepatocellular carcinoma. Methods: The interaction of Lin28 and circRNA was predicted by circinteractome database. The binding of Lin28 to the selected gene was determined by RIP and qRT-PCR. Construction of circular RNA overexpression vector by circular RNA lentiviral expression vector PLO-ciR. qRT-PCR was used to detect the overexpression of circRNA in hepatocellular carcinoma cell lines. The expression of Lin28 protein in hepatocellular carcinoma cell line was detected by Western blot. Results: Lin28 interacted with circRNAs. circRNA overexpression vectors were successfully constructed. circRNAs were successfully overexpressed in hepatocellular carcinoma cell lines. The expression of Lin28 protein in the circRNA overexpressing hepatocellular carcinoma cell lines was decreased. Conclusion: circRNA plays an important role in the regulation of Lin28 expression in hepatocarcinoma cell lines.
