长链非编码RNA H19通过MiR-675 调控膀胱癌发生发展的分子机制研究

摘要:通过探究H19/miR-675及其靶基因在膀胱癌发生、发展中的相互作用,阐释H19在膀胱癌中发挥作用的机制。采用人膀胱尿路上皮癌细胞系(T24,5637,EJ)以及人正常膀胱上皮细胞系(SV-HUC-1),利用实时荧光定量PCR (SYBR Green Ⅰ法)检测细胞及组织中H19, miR-675以及RB基因表达情况,利用Western-blot检测细胞和组织中RB蛋白的表达水平。采用膀胱尿路上皮癌细胞系T24,转染H19的过表达pc-DNA及siRNA,检测miR-675的表达水平以及膀胱癌细胞增殖和迁移能力; 转染miR-675的mimics及inhibitor,检测RB蛋白的表达情况以及膀胱癌细胞增殖和迁移能力;转染RB基因pc-DNA及siRNA,检测膀胱癌细胞增殖和迁移能力。结果显示, H19,miR-675在膀胱癌细胞中的表达水平显著上调(p<0.05),与正常膀胱上皮组织相比,在膀胱癌组织中H19, miR-675的也显著高表达,两者差异有统计学意义(p<0.05)。过表达H19基因会导致miR-675表达量的显著上调,同时促进T24细胞的增殖和迁移;而质粒干扰H19表达则导致miR-675表达量的显著下调,同时抑制T24细胞增殖和迁移。证实了在膀胱癌中存在H19/miR-675/RB调控通路。

Role of lncRNA H19 derived miR-675 in promoting bladder cancer cell proliferation and progression
Abstract:To detect the expression level of H19, miR-675 and RB in bladder cancer cell lines and tissues adn verify the function of H19/miR-675 in promoting bladder cancer proliferation through down regulating their target gene. we included 3 types of bladder cancer cell lines (T24, 5637, EJ) and normal bladder cell line (SV-HUC-1). Real-time PCR (SYBR Green Ⅰ) was performed to detecting the expression level of Role of lncRNA H19 derived miR-675 in promoting bladder cancer cell proliferation and progressionH19, miR-675 and RB in bladder cancer cell lines and tissues, while Western-blot were performed for detecting the expression level of RB protein in cells and tissues. we selected T24 cell line as study subject, and then transfected T24 cell with H19 pc-DNA and siRNA, detecting the expression level of miR-675 and the ability of cell proliferation and migration; transfected T24 cell with miR-675 mimics and inhibitor, detecting the expression level of RB protein and the ability of cell proliferation and migration; transfected T24 cell with RB pc-DNA and siRNA, detecting the ability of cell proliferation and migration. The results showed that, comparing to normal bladder cell line, the expression levels of H19 and miR-675 are significant increased in bladder cancer cell lines. Meanwhile, comparing to normal bladder tissue, the expression levels of H19 and miR-675 are increased in bladder cancer tissue, the differences had significances (p<0.05). The expression level of RB in bladder cancer tissue was decreased while increased in normal bladder tissue. In this experiment, we have confirmed that the H19/miR-675/RB pathway functions vitally in the formation and developing of the bladder cancer.
