脑微血管内皮细胞中Atg7条件性敲除小鼠的制备及其脑微血管分布观察

摘要:目的构建脑微血管内皮细胞中Atg7条件性敲除小鼠(Atg7-/-)及观察脑微血管的分布。方法收集小鼠鼠尾样本,试剂盒提取鼠尾DNA,设计并合成相关引物,PCR及琼脂糖电泳分析小鼠基因型。收集1月及3月龄小鼠大脑进行冰冻切片,免疫荧光检测野生型及Atg7-/-小鼠的血管标记物的分布。结果 Atg7-/-小鼠构建成功。1月龄Atg7-/-小鼠与野生型小鼠相比,脑皮质血管密度没有明显差别。此外,3月龄Atg7-/-小鼠与野生型小鼠相比,脑皮质血管密度显著减少结论脑微血管内皮细胞中Atg7的敲除降低脑皮质血管的密度。

Preparation of Atg7 conditional knockout mice in brain microvascular endothelial cells and observation of cerebral microvascular distribution
Abstract:Aim Construction of Atg7 conditional knockout mice (Atg7-/-) in brain microvascular endothelial cells and observation of the distribution of cerebral microvessels. Method The mouse tail samples were collected and the mouse tail DNA was extracted by the DNA extraction Kit. The relevant primers were designed and synthesized. PCR and agarose gel electrophoresis were used to analyze the genotypes of mice. The brains from two months and nine months aged mice were collected and frozen sections were used to detect the distribution of vascular markers in wild-type and Atg7-/- mice. Results Atg7-/- mice were successfully constructed. There was no significant difference in cerebral cortical blood vessel density between one months of Atg7-/- mice and wild-type mice. In addition, compared with wild-type mice, the density of cerebral cortical vascular of three months aged Atg7-/- mice was significantly decreased. Conclusion The knockout of Atg7 in cerebral microvascular endothelial cells can decrease the density of cerebral cortical vascular.
