摘要:目的:研究高表达RHBDF1对MCF-10A乳腺腺泡3D形态发生的影响。方法:采用乳腺细胞3D培养和Matrigel Plug的方法模拟乳腺腺泡发生过程,用免疫荧光和Western Blot的方法检测腺泡发生过程中细胞凋亡蛋白Cleaved Caspase-3、Bim2和细胞增殖蛋白Ki67和Survivin的表达及Akt的磷酸化;用H&E染色观察RHBDF1在小鼠体内对乳腺腺泡的发生的影响。结果:RHBDF1通过抑制Cleaved Caspase-3、Bim2,上调Ki67和Survivin的表达及Akt的磷酸化破坏乳腺腺泡的发生。结论:过表达RHBDF1能够破坏乳腺腺泡的发生,其可作为一个潜在的乳腺癌治疗的新靶标。

RHBDF1 Overexpression Inhibits Apoptosis of MCF-10A to Perturb Acini Morphogenesis in 3D Cultures
Abstract:Objective: To investigate the role of RHBDF1 overexpression in MCF-10A to form morphological phenotype and hollow lumen formation of mammary acini in 3-D cultures. Methods: Immunostaining and western blot were employed to detect the protein expression level of cleaved Caspase-3, Bim2, Ki67, Survivin, and phosphorylated Akt in mammary acini during morphogenesis by 3D culture. In addition, H&E staining was employed to examine the mammary acini during morphogenesis in NOD/SCID mice by Matrigel plug. Results: RHBDF1 can perturbs morphological phenotype and hollow lumen formation of mammary acini by downregulate the protein expression level of cleaved Caspase-3, Bim2 and upregulate the protein expression level of Ki67, Survivin, and phosphorylated Akt in vitro and in vivo. Conclusion: RHBDF1 overexpression in MCF-10A perturbs morphological phenotype and hollow lumen formation of mammary acini, and it may be as a novel target in the treatment of breast cancer.
