宫内砷暴露对雄性子代小鼠精子质量的影响

摘要:目的:探索宫内无机砷暴露对雄性子代小鼠生殖功能相关指标的影响。方法:将F0代孕鼠随机分为砷处理组和对照组,每组8只。在GD8-18天通过自由饮水方式对孕鼠进行染毒处理,其中砷处理组饮水中亚砷酸钠浓度为42.5 mg/L。待F1代仔鼠出生后测量雄性子代小鼠的肛殖距、体重等指标。F1代子鼠饲养至90天,颈椎脱臼处死,取双侧睾丸和附睾组织并称重,同时镜下观察精子活动度和畸形。结果:两组小鼠相比,砷处理组小鼠体重(38.59 ± 2.02 g)高于对照组小鼠(32.62 ± 4.15 g),且差异具有统计学意义(P < 0.05)。对照组和砷处理组小鼠相比,肛殖距长度和睾丸重量无显著差异(P > 0.05)。而对于双侧睾丸脏器系数,砷处理组小鼠较对照组小,且差异具有统计学意义(P < 0.05)。精子活力观察结果表明,砷处理组小鼠精子活力(85.37 ± 4.52%)显著低于对照组小鼠(91.07 ± 3.52%)(P < 0.05)。精子畸形结果表明,砷处理组小鼠精子总畸形率(9.58 ± 2.60%)显著高于对照组小鼠精子总畸形率(5.55 ± 1.65%)(P < 0.05)。结论:宫内砷暴露可以引起雄性子代精子质量降低。

Effect of in Utero Arsenic Exposure on Sperm Quality of Male Offsprings
Abstract:The study aims to explore the effects of in utero arsenic exposure on the rEffect of in Utero Arsenic Exposure on Sperm Quality of Male Offspringseproductive parameters of male offspring mice. F0 pregnant mice were randomly divided into control and arsenic treatment groups. There are 8 mice in each group. Animals were treated with either deionized water or water containing 42.5 mg/L of sodium arsenite. The control mice received deionized water throughout the life. The arsenic treatment mice received deionized water containing 42.5 mg/L of sodium arsenite from GD8 to 18. Mice were monitored daily for signs of toxicity and discomfort. All dams were allowed to deliver naturally and to rear their offsprings to weaning until PND 21. The litters of each group were culled on PND 4 to keep litter size at 8-10 pups/litter. The F1 mice were observed daily and recorded their body weight and anogenital distance weekly. Male offspring mice were sacrificed on PND 90. Samples of testis and epididymis tissues were collected. Sperm quality was determined by activity and malformation. Body weight was significantly higher in mice exposed to arsenic than that in controls (38.59 ± 2.02 g VS 32.62 ± 4.15 g). There are no difference in anogenital distance and testicular weight between arsenic treatment mice and the control mice. However, the organ coefficient of testes was significantly lower in mice exposed to arsenic than that in controls. The viability of epididymal spermatozoa in mice was 91.07 ± 3.52% in control group and 85.37 ± 4.52% in arsenic treatment group. It indicates that arsenic treatment mice had a lower viability of epididymal spermatozoa than that in control mice. Meanwhile, sperm malformation rate increased from 5.55 ± 1.65% in controls to 9.58 ± 2.60% in arsenic treatment group. In utero arsenic exposure may have reproductive toxicity on male offspring.
