SOX5调控MITF-M影响黑色素细胞色素生成

摘要:本研究旨在探讨SOX5在不同毛色小鼠皮肤是否存在差异表达,以及与毛色形成是否存在相关性。免疫荧光结果显示SOX5在不同毛色小鼠皮肤中均可以表达,且其主要表达部位为毛囊外根鞘。QRT-PCR检测显示SOX5在黑色和灰色小鼠皮肤中mRNA相对表达量高于棕色小鼠皮肤;蛋白免疫印迹结果表明SOX5在黑色和灰色小鼠皮肤中蛋白水平高于棕色小鼠皮肤。为了进一步研究SOX5在黑色素细胞色素沉着的重要作用,我们把SOX5转染到小鼠黑色素细胞并检测色素沉着相关基因的表达水平,同时测定黑色素含量。结果表明转染SOX5与空载组相比MITF-M、TYR 、TYRP1、TYRP2、PMEL、OA1表达量均升高,黑色素含量也明显增加。同时通过过表达MITF-M发现MITF-M对SOX5存在负反馈作用。SOX5在不同毛色小鼠皮肤中均可有效表达,且差异显著。以及SOX5过量表达使色素沉着相关基因的表达量及黑色素含量增加。由此表明SOX5通过调控MITF-M影响色素的生成进而参与毛色的形成。

Effects of SOX5 regulates MITF-M on Melanogenesis in Mouse Melanocytes
Abstract:The aim of this study was to investigate whether SOX5 has different expression in mouse skin with different hair color and whether there is a correlation with the formation of coat color. Immunofluorescence results showed that SOX5 could be expressed in different coat color mouse skin, and its main expression site was hair follicle outer root sheath. QRT-PCR assay suggested that the relative expression of SOX5 mRNA in black and gray mouse skin was higher than that in brown mouse skin. The results of immunoblotting showed that the protein level of SOX5 in skin of black and gray mice was higher than that of brown mouse skin. In order to further study the important role of SOX5 in melanocyte pigmentation, we transfected SOX5 into mouse melanocytes and detected the expression level of the pigmentation-related genes, and measured the melanin content. The results showed that the expression levels of MITF-M, TYR, TYRP1, TYRP2, PMEL and OA1 were increased and the content of melanin was significantly increased in SOX5 transfected group, compared with empty vector group. And it was found that MITF-M had a negative feedback on SOX5 by overexpression of MITF-M. SOX5 can be expressed effectively in mouse skin with different hair color, and the difference is significant. And overexpression of SOX5 increased the expression level of the pigmentation-related genes and the content of melanin. These suggest that SOX5 affected the formation of pigments and then involved in the formation of coat color by regulating MITF-M.
