鼎湖鳞伞菌丝体多糖免疫活性研究

摘要:药用真菌对人体健康具有多种积极作用,其中的多糖成分具有免疫调节活性。本实验采用DEAE Sepharose Fast Flow和Sephadex G-100柱层析得到鼎湖鳞伞(Pholiota dinghuensis Bi)菌丝体多糖(PDP)的三个组分PDP-1、PDP-2和PDP-3。通过RAW264.7细胞实验研究了粗PDP及纯化组分PDP-1、PDP-2、PDP-3的体外免疫调节活性。结果显示在各个多糖组分中,PDP-2和PDP-3表现出较好的体外免疫促进作用,有效地促进了RAW264.7细胞的增殖,并显著提高其吞噬中性红能力及酸性磷酸酶活性。采用环磷酰胺免疫抑制小鼠模型,探讨了PDP-2的体内免疫调节活性及其协同抗氧化作用。结果表明,PDP-2能明显提高环磷酰胺免疫抑制小鼠的脾脏指数、血清溶血素水平及血清、脾脏中的溶菌酶含量,还能协同增强小鼠肝脏中T-AOC、T-SOD、CAT和GSH-Px 的酶活,降低MDA 的水平。因此,PDP具有一定的免疫调节活性。

Study on the Immune Activity of Pholiota dinghuensis Bi mycelia polysaccharides
Abstract:Medical mushrooms have positive effects on human health, and their polysaccharides possess immunomodulatory activities. In the present study, DEAE Sepharose Fast Flow and Sephadex G-100 column chromatography were utilized to purify the crude polysaccharides from Pholiota dinghuensis Bi mycelia (PDP) to afford three fractions of PDP-1, PDP-2 and PDP-3. The immunomodulatory potentials in vitro of crude PDP and its purified fractions on RAW264.7 cell line were investigated. The data indicated that PDP-2 and PDP-3 could significantly stimulate the proliferation of macrophages, enhance the capability of macrophage phagocytosis and increase the activity of acidic phosphatase. The cyclophosphamide-induced immunosuppression mice model was further used to evaluate the immunomodulatory activity of PDP-2. It was found that PDP-2 could increase the spleen index, hemolysin level, lysozyme content and the activities of T-AOC, T-SOD, CAT, GSH-Px, and reduce the MDA level. The results suggested that PDP had immunomodulatory activities.
