8-十六烷基小檗碱抗肿瘤作用的研究

摘要:为研究小檗碱衍生物8-十六烷基小檗碱对人A549 肺癌细胞的增殖抑制作用和对荷瘤小鼠模型肿瘤生长抑制作用。采用MTT法检测8-十六烷基小檗碱对人A549 肺癌细胞增殖的抑制作用;BALB/c 裸鼠右前肢皮下接种A549 细胞后随机分组,灌胃不同剂量[5、10 mg/(kg.d)]的8-十六烷基小檗碱以及小檗碱[120 mg/(kg.d)],计算小檗碱及8-十六烷基小檗碱对A549裸鼠移植瘤模型实体瘤的抑制率。结果表明8-十六烷基小檗碱对A549 有较好地抑制作用(IC50= 2.3 μg/ml),相比小檗碱(IC50= 27.4 μg/ml)抑制作用提高近10倍;8-十六烷基小檗碱能有效地抑制A549裸鼠移植瘤的生长,低、高剂量的抑制率分别为34.58%、59.07%;

Studies on the Inhibiting Tumor Effect of 8-Cetylberberine
Abstract:To explore the anti-tumor activity of 8-cetylberberine on human A549 lung cancer cell and its xenograft mice. Inhibition of 8-cetylberberine on A549 was determined with an MTT assay. A549 xenograft mice were administered orally with 5, 10 mg/(kg.d) dosage of 8-cetylberberine and 120mg/(kg.d) dosage of berberine, then the tumor inhibition rate were measured. The result showed that 8-cetylberberine exhibited more significantly potent than BBR, with a ten times lower IC50 than that of BBR after 24 h treatment (2.3 vs 27.4 μg/mL); and 8-cetylberberine at a dose of 10 mg/kg could significantly suppress tumor growth in A549 xenograft mice, which were even stronger than those of BBR treatment at a dose of 120 mg/kg.
