表达PyMSP9蛋白的重组腺病毒疫苗构建及其免疫原性检测

摘要:疟原虫裂殖子表面蛋白9(MSP9)是一个潜在的疟疾疫苗候选抗原,但目前对于MSP9的全长蛋白的免疫原性研究还未见报道。本文利用人5型腺病毒载体构建表达约氏疟原虫裂殖子表面蛋白9(PyMSP9)的重组腺病毒疫苗,研究该疫苗在小鼠体内的免疫原性。结果显示,本文成功构建了表达PyMSP9蛋白的重组腺病毒疫苗,并且将该疫苗免疫小鼠,免疫后获得的免疫血清能有效识别重组蛋白PyMSP9-Nt片段。该结果表明表达PyMSP9蛋白的重组腺病毒疫苗能有效激活小鼠的免疫反应,具有一定的免疫原性,为MSP9疫苗的开发提供理论基础。

Construction of Human adenoviral recombinant vector expressing P.yoelii merozoite surface protein 9 and its immunogenicity detection
Abstract:Malaria parasite merozoite surface protein 9(MSP9) is one of the malaria vaccine candidate, but the immunogenicity study of full MSP9 protein is not reported. In this paper,Human adenoviral recombinant vector expressing P.yoelii merozite surface protein 9 is constructed, and its immunogenicity is detected in mice. The result shows that Human adenoviral recombinant vector expressing PyMSP9 is successfully prepared, and the mice immune serum can recognize the recombinant PyMSP9-Nt protein. It indicates that Human adenoviral recombinant vector expressing PyMSP9 can induce the mice immune response, and its immunogenicity is successfully detected. It provide the theoretical basis for the development of MSP9 vaccine.
