葡萄糖苷酶的化学修饰及其对酶催化性能的影响

摘要:以聚乙二醇衍生物及葡萄糖苷酶的化学修饰及其对酶催化性能的影响柠康酸酐为修饰剂,对葡萄糖苷酶进行化学修饰,考察了不同修饰剂和投料比对修饰酶催化活性的影响,通过SDS-PAGE对修饰酶进行表征。实验结果表明:柠康酸酐修饰后的酶催化活性大大降低,而通过聚乙二醇类衍生物修饰后的酶催化活性及稳定性有了一定的改善。

Chemical modification of glucosidase and its effect on enzyme catalytic performance
Abstract:The effects of different modifiers and feed ratio on the catalytic activity of the modified enzyme were investigated by using polyethylene glycol derivative and citraconic anhydride as modifier. The modified enzyme was characterized by SDS-PAGE. The results showed that the catalytic activity of citraconic anhydride was greatly reduced, and the activity and stability of the enzyme modified by polyethylene glycol derivatives were improved.
