白姜花EIL1基因的克隆及生物信息学分析

摘要:以白姜花(Hedychium coronarium)为材料,利用RT-PCR技术克隆得到两个乙烯途径的转录因子EIL1基因,分别命名为HcEIL1-1和HcEIL1-2。对这两个基因的氨基酸序列进行结构域分析,得出在N端都存在两个EIN3-like protein DNA结合结构域;和其他植物EIN3/EILs进行系统进化树分析和同源性分析,得出这两个基因与马来西亚野生香蕉EIL1亲缘关系最近;利用核定位预测软件分析,得出这两个基因都存在核定位信号;利用亲水性分析软件分析了这两个基因蛋白各个部位的亲水性情况;利用蛋白高级结构预测软件对这两个蛋白进行了二级结构和三级结构分析,预测并构建了这两个基因的蛋白三维结构图。

Cloning and Functional Bioinformatics Analysis of EIL1 Gene from Hedychiumcoronarium
Abstract:two transcription factor genes of ethylene pathway in H.coronarium were cloned by RT-PCR, and named as HcEIL1-1 and HcEIL1-2. The amino acid sequences of these two genes were analyzed by domain analysis, and two EIN3-like protein DNA binding domains are found at the N-terminus; We analyzed HcEIL1-1 and HcEIL1-2 with other plants EIN3/EILs by phylogenetic tree analysis and homology analysis, and results show that these two genes are closely related to MaEIL1(Musa acuminata subsp. Malaccensis);it is concluded that both genes have nuclear localization signals by using nuclear localization prediction software;and we analyzed hydrophilicity of the two gene proteins by hydrophilicity analysis software. protein advanced structures of the two genes were predicted and constructed by using the protein advanced structure prediction software.
