摘要:大豆MDH1蛋白为NAD+/NADH依赖型苹果酸脱氢酶(Malate dehydrogenase, MDH, EC 1.1.1.37),这是一种古老的氧化还原酶,在细菌,古细菌及真核生物中广泛存在。本实验通过在洋葱表皮及烟草叶片中瞬时表达MDH1-GFP,确认了MDH1定位于线粒体,并发现其N末端第二的氨基酸对其正确定位十分重要。本实验还制备并纯化了MDH1的特异性多克隆抗体,为今后研究MDH1的作用机制奠定了基础,也为检测植物线粒体MDH水平进一步研究其作用机制提供了有用的工具。

Sublocalization of malate dehydrogenase 1 in soybean and its antibody preparation
Abstract:Soybean MDH1 is a NAD+/NADH dependent malate dehydrogenase (MDH, EC 1.1.1.37), which reversibly catalyzes the conversion of malate to oxaloacetate and exists in almost all living organisms including eubacteria, archea, and eukaryotes. Results from present study verified that soybean MDH1 is localized in mitochondria through transient expressing MDH1-GFP in both onion epidermal cells and the leaves of Nicotiana benthamiana. It is also revealed that the second amino acid is essential for transporting MDH1 into mitochondria. The polyclonal antibody prepared in this study is highly efficient and specific, which provides a useful tool to study the mechanism of plant mitochondria MDH.
