摘要:目的探究miR-223能否通过调控人血管平滑肌细胞自噬功能促进其迁移功能。方法收集正常捐献者的下肢动脉标本并提取HASMCs,通过Transwell小室实验、划痕损伤愈合实验与细胞免疫荧光实验观察比较实验组(转染miR-223 mimic)和阴性对照组两组细胞迁移功能差异,并比较自噬相关蛋白表达差异。结果转染miR-223 mimic的HASMCs与阴性对照组相比,实验组迁移能力明显增强,并可被自噬抑制剂3-MA抑制。细胞免疫荧光显示实验组自噬功能明显增强。结论 miR-223可通过提高自噬水平促进HASMCs迁移功能。?????

MicroRNA-223 promotes the migration of vascular smooth muscle cells by regulating autophagy
Abstract:Purpose To explore the role of miR-223 on the regulation of migration function of human vascular smooth muscle cells, through autophagy flux. Methods Donors\' normal lower limb arteries were collected and HASMCs were extracted. By Transwell chamber, Wound Healing and cell immunofluorescence, differences of cells migration function and autophagy related protein expression between experimental group (miR-223 mimic transfected) and negative control group were observed. Results The migration capacity of the HASMCs transfected with miR-223 mimic group was significantly enhanced with an enhancement of autophagy flux, which was down-regulated by the autophagy inhibitor 3-MA. Conclusion miR-223 up-regulates HASMCs\' autophagy flux, and promoting the migration function of HASMCs.
