乳腺癌细胞在三维细胞外基质中的迁移模型

摘要:细胞外基质中的胶原蛋白纤维会对附近细胞的形态、运动产生关键作用。本文建立了剪切流动下的计算模型,以模拟乳腺癌细胞在细胞外基质中的迁移。间隙液流动会对蛋白纤维排列产生影响,亦会改变细胞周围自趋化物质的浓度场。蛋白纤维的排列方式是肿瘤细胞迁移的重要影响因素。计算结果表明,在剪切流动下,肿瘤细胞的迁移方向与流动方向相关,而且流速越大,相关性越高。该结论与其他实验研究结果相一致,证明了本文的计算模型能够合理地预测乳腺癌细胞在三维细胞外基质中的转移。

A model for a breast cancer cell migrating in 3D ECM
Abstract:Collagen fibers of the extracellular matrix play a key role for regulating the morphology and migration of the surrounding cells. In this paper, a model involved in the shear flow is established to simulate the breast tumor cell migration in an extracellular matrix. The interstitial flow affects not only the collagen fiber alignment but also gradient of the chemotaxis kinase. The collagen fiber alignment is important to cell migration. Results show that the direction of cell migration is related to that of the flow. Moreover, the consistency of these two directions goes up with the flow speed. The simulation results agree well with the previous experimental findings and indicate that this model can better predict migration of tumor cells in 3D extracellular matrix.
