牡丹不同部位提取物多酚黄酮含量及抗氧化活性比较

摘要:【目的】为充分开发牡丹在抗氧化剂、医药保健及相关领域的研究和应用提供一定理论和实验基础。【方法】以牡丹花瓣、花粉、种皮、根皮为材料,测定了各部位中总酚、总黄酮的含量,后以VC、VE、没食子酸丙酯(PG)、2,6-二叔丁基-4-甲基苯酚(BHT)为对照物,选用还原能力、DPPH自由基清除率、ABTS自由基清除率及在β-胡萝卜素/亚油酸自氧化体系中的抗氧化活性系数(AAC)为指标对不同部位醇提物进行抗氧化活性评价。【结果】牡丹各部位中总酚、总黄酮含量高低顺序皆为:种皮>花粉>根皮>花瓣;在浓度为1.0mg/mL时,各物质还原能力强弱顺序为:VE≥VC≥PG≥BHT>花瓣≥根皮>种皮>花粉;DPPH自由基清除能力强弱顺序为:PG>VC>VE>根皮>花瓣>BHT>种皮≥花粉;清除ABTS自由基能力顺序为:VC≥VE≥PG≥BHT>种皮>花瓣≥花粉>根皮;AAC大小顺序为:VE≥种皮≥BHT≥PG>花瓣≥根皮>VC>花粉。牡丹各部位抗氧化活性均随着浓度增大而增强,总酚、总黄酮与DPPH清除能力呈负相关;总酚、总黄酮与ABTS清除能力呈现良好正相关关系;对牡丹花瓣中黄酮组成分析,发现主要为山柰酚、木犀草素、芹菜素的糖苷。【结论】牡丹各部位均具有一定的抗氧化能力,花瓣提取物在各个指标中抗氧化性能均较强,可能成为一种新型高效的天然抗氧化剂。

Comparative on polyphenol,flavonoid and antioxidant activities from different parts of Paeonia suffruticosa Andr.
Abstract:Objective: To provide a theoretical and experimental basis for the full development of peony in the research and application in antioxidants, medical care and related fields.Methods: Peony petal, pollen, seed coat and root bark were used to determine the contents of polyphenols and flavonoids in different parts of the plant. VC, VE, PG and 2,6-di-tert-butyl-methylphenol (BHT) as control, reducing power, DPPH radical scavenging rate, ABTS free radical scavenging rate and antioxidant activity coefficient (AAC) in β-carotene / linoleic acid autoxidation system Antioxidant activity of alcohol extracts from different parts was evaluated.Results: Polyphenols, flavonoids in various parts of peony all were in the order of the seed coat> pollen> bark> petal. At the concentration of 1.0 mg / mL, the order of the reducing ability of each substance was VE≥VC≥PG≥BHT>petal≥ bark>seed coat> pollen; DPPH radical scavenging ability was in the order of PG> VC> VE> root bark> petal> BHT> seed coat ≥ pollen; ABTS free radical scavenging ability order was VC≥VE≥PG≥BHT> seed coat> petal≥ pollen> root bark; AAC value was in the order of BHT> PG> petals> root bark> VC> pollen. Antioxidant activity of all parts of peony was increased with the concentration. Polyphenols and flavonoids content were negatively correlated with DPPH scavenging ability. Polyphenols and flavonoids showed a good positive correlation with ABTS scavenging capacity. Analysis of the composition of the flavonoids in the petals of peony, found mainly are kaempferol, luteolin, apigenin and its glycosides.Conclusion: Parts in Peony all have a certain antioxidant capacity, Petal extract had a strong antioxidant performance in all indicators. So petal may become a new kind of highly effective natural antioxidants.
