产磷脂酶D菌株的筛选及其发酵优化研究

摘要:经过平板初筛和摇瓶复筛,从土壤中筛选出了7株产磷脂酶D菌株,对其中一株酶活较高的菌株进行菌落形态观察,生理特征鉴定,16S rRNA序列分析。经鉴定其为肉桂链霉菌,命名为Streptomyces cinnamoneus SK43.003。通过摇瓶发酵培养条件和培养基组分优化研究,酶活可达7.3U/ml。在以卵磷脂为底物及丝氨酸存在的条件下,利用该磷脂酶D进行两相反应可转化生成磷脂酰丝氨酸,转化率为92%。

Screening and fermentation study of a phospholipase D-producing strain
Abstract:Seven strains producing phospholipase D were isolated from soil through the screening of yolk medium plate and shake flask culture. It was identified as streptomyces by colony morphology, physiological characteristics and sequence analysis of 16S rRNA and named as Streptomyces cinnamoneus SK43.003. Fermentation conditions and optimum medium of PLD production were optimized, in which the enzyme activity reached 7.3U/ml. In the presence of serine, the lecithin was convert to phosphatidylserine with a 92% transformation rate in a biphasic system.
