丁酸梭菌对小鼠急性胰腺炎及相关肠道炎症的保护作用研究

摘要:目的探讨丁酸梭菌对小鼠急性胰腺炎及相关肠道炎症的保护作用及机制。方法 将Balb/c小鼠随机分为正常对照组、急性胰腺炎模型组和Clostrimdium Butyricum CGMCC0313.1(CB0313.1)干预组。连续21天给予干预组小鼠CB0313.1,在第22天时,建立雨蛙素诱导的急性胰腺炎模型。采用商业试剂盒测定小鼠血清淀粉酶、脂肪酶及胰腺髓过氧化物酶活力;采用酶联免疫法测定胰腺和结肠的炎症因子水平;采用流式细胞术检测胰腺中性粒和树突细胞的浸润情况;采用蛋白免疫印迹法检测胰腺和结肠NLR热蛋白结构域亚家族成员3(NLRP3)及核因子(NF)-κB信号通路。结果 与模型组相比,CB0313.1干预组小鼠的血清淀粉酶、脂肪酶及胰腺髓过氧化物酶活力显著降低;胰腺水肿及中性粒和树突细胞浸润得到显著改善;胰腺和结肠炎症因子水平显著下调;胰腺和结肠NLRP3炎症小体及NF-κB的激活受到抑制。结论 CB0313.1预处理对小鼠急性胰腺炎及相关肠道炎症具有保护作用,其作用机制与缓解胰腺中性粒和树突细胞浸润,抑制胰腺和结肠NLRP3及NF-κB介导的炎症反应有关。

Protective efficacy of Clostrimdium Butyricum on acute pancreatitis and associated intestinal inflammation in mice
Abstract:Objective To investigate the effects of Clostrimdium Butyricum on acute pancreatitis (AP) and associated intestinal inflammation and explore the potential mechanism. Methods Balb/c mice were randomly divided into control group, AP group, and Clostrimdium Butyricum CGMCC0313.1 (CB0313.1) prophylactic group. The mice in intervention group were treated with CB0313.1 once a day for 21 consecutive days. AP was induced with cearuline on 22st day. The levels of serum amylase and lipase and pancreatic myeloperoxidase (MPO) were determined by relevant commercially available kits, respectively. Pancreas and colon were assayed for inflammatory cytokine using a sandwich ELISA kit. The infiltration of neutrophils and dendritic cells (DCs) in pancreas were examined by flow cytometer. Expression of NOD-like receptor family protein 3 (NLRP3) inflammasome and nuclear factor (NF)-κB in pancreas and colon were determined by western blot. Results Compared with AP group, the mice in CB0313.1 prophylactic group presented significantly reduced serum amylase and lipase levels and pancreatic myeloperoxidase activity, as well as decreased edema and infiltration of neutrophils and DCs in pancreas. The protective effect was also confirmed by decreased inflammatory cytokine in pancreas and colon. Furthermore, CB0313.1 suppress NLRP3 and NF-κB signaling pathway in pancreas and colon. Conclution CB0313.1 protect experimental acute pancreatitis and related intestinal inflammation by improving infiltration of neutrophils and DCs and inhibiting inflammation mediated via NLRP3 inflammasome and NF-κB.
