气相色谱-质谱联用法同时分析细胞内17种氨基酸

摘要:建立了一种基于气相色谱-串联质谱技术(GC-MS/MS)的氨基酸快速分析定量方法。细胞样品采用80%甲醇溶液提取,BSTFA+1%TMCS衍生后,用GC-MS/MS进行定量分析,分析时间为27 min。该分析方法的线性范围为0.5~200 mg/L,方法检出限在0.01~0.68 mg/L之间, 加标回收率为77. 0%~118.6%,相对标准偏差为1.5%~10.4%,可对细胞中17种氨基酸成分进行定量分析。本方法具有处理过程简便、快速、灵敏度高等特点,适用于哺乳动物细胞等样品中氨基酸类代谢产物的快速分析。

Rapid Analysis of Seventeen Amino Acids in Mammal Cells by Gas Chromatography-Tandem Mass Spectrometry
Abstract:A rapid quantitative method using gas chromatography-tandem mass spectrometry (GC-MS/MS) was developed for the analysis of amino acids in mammal cells. Cells were extracted by 80% methanol, derivated by BSTFA+1%TMCS, and then analyzed by GC-MS/MS. A total of 17 amino acids were detected and the data were acquired in selective reaction monitoring (SRM) mode. The method showed a good linearity between 5~200 mg/L with a inter-day precision of 1.5% ~ 10.4% (RSDs). And limits of detection (LODs) and recoveries of the method were 0.01~0.68 mg/L and 77. 0%~118.6%, respectively. These results indicated that the method is simple, rapid, highly sensitive, and is suitable for the detection of amino acids in mammal cells.
